It is with regret that with this letter I tender my resignation from your employment, and are therefore giving up my post of Sales Assistant, observing two weeks’ notice from the date of the letter as per the terms and conditions of my contract of employment. Should you wish to forfeit this period of worked notice I would be willing to exchange the period for paid absence.

I have enjoyed my time with you and would like to thank you for the skills and experience you have furnished me with during my employment. I would be happy to assist in the procurement and training of a replacement employee should you wish me to. I will complete my two weeks with the same attention and level of service as always, or should you prefer, I am happy to accept the two weeks notice as paid absence.

I hope that you take this resignation with the good will in which it is issued. I hope we may once again work together in the future.

Not a bad letter, I would change it slightly as some of the grammar is incorrect (well, I feel it is, sorry ) for example where you have written
 
"It is with regret that with this letter I tender my resignation from your employment, and are therefore giving up my post of Sales Assistant, observing "
 
I would actually put
 
It is with regret that with this letter I tender my resignation from your employment, and therefore I am giving up my post of "  it just reads better to me
